In a word, Yes.  Buna-N (or nitrile) will resist transmission fluid and is
readily available.  But EPDM is commonly sold for plumbing applications and
transmission fluid/oil will slowly dissolve it.  There are lots of more exotic
materials available, but Buna-N should do fine.
