White spirit should do the job, if it is proper mastic at least. Won't
harm the paint, but it tends to be a somewhat messy job, so it pays off
to get much of it off by mechanical means first (by use of a blunt
plastic or wood scraper, for instance).

Rub the seal with washing up liquid first, that makes all the difference
when it comes to a smooth installation.

FWIW, the traditional window rubber seals as used on Triumphs *are*
designed to be sealed off with mastic. IMHO that is also the only way of
getting them properly water proof, as well as avoid any change of water
collecting along the edge and starting rust.
But DO ensure that you use the proper non-curing mastic for automotive
use - and stay well clear of any silicone sealer or similar. Excess
mastic is removed by use of white spirit.
